Glucosamine is a fundamental building block and required for the synthesis of glycosaminoglycan, proteoglycan and hyaluronate, which are important for collagen synthesis in cartilaginous tissue, tendons and ligaments, degradation of which can lead to osteoarthritis (OA). Moreover, glucosamine sulphate exhibits anti-inflammatory and chondroprotective actions and thus has been shown to alleviate pain and even restore or stimulate articular cartilage formation in patients with damaged cartilages in affected joints with OA. Chondroitin sulphate is a natural glycosaminoglycan, present in the extracellular matrix of the resilient cartilage tissues. Apart from that, it reduces the formation of pro-inflammatory cytokines and pro-inflammatory enzymes, such as phospholipase A2, cyclooxygenase 2 (COX- 2) and nitric oxide synthase. It also exhibits chondroprotective properties. Combined action of glucosamine sulfate and chondroitin sulphate has been shown to delay radiological progression of OA. Ginger Extract contains 6-shogaol, the active constituent, who has an inhibitory effect on the release of substance P, a neuropeptide associated with inflammation and pain. Thus, it modestly improves pain and disability in OA. Vitamins and minerals provide antioxidant action to protect the cell damage and supply necessary cofactors, which are the key components in enzymatic reaction for the synthesis of cartilage tissues. Vitamin D is required for the regulation of calcium and phosphorus which maintains the proper bone structure. Vitamin E is an antioxidant having small pain killing activity and complements the effects of other anti-inflammatory ingredients.<\/p>\n
